Wisconsin | Yes, it will make a big difference. Even better if you use canned expanding foam around the edges of the polyiso, and the plywood so the walls are air tight. Along with the ceiling and any other joints. Cut the foam 1/2-3/4" short and fill the gap works good. You'll never eliminate big door air leakage, but if the rest isn't leaking it will limit the amount of heat lost through the big door.
