Shy or introverted people generally don't like being the center of attention obviously, so it's a lot of mental prep ahead of time.

Avoid stress ahead of time, so tell him to do the most relaxing or thing he enjoys before.
Avoid as many interactions with people ahead of time. Shy people might find things social exhausting to begin with, so avoid extra mental stress by just avoiding people as much as possible beforehand.
Have him practice talking about his skills and/or accomplishments out loud ahead of time, even if it's in a mirror. That will at least get him more comfortable hearing himself talk aloud, plus repetition.
Most obvious things are also the hardest. Eye contact, sit up straight, firm handshake if offered.
Might not even be a bad idea for him to tell the interviewer that he has some trouble with social interaction, but that he's working on it.
You can tell him that life is full of difficult things, but we all have to do them. And worst case he won't get the offer, but if he doesn't try he'll never know if he could have.
If they trip him up and ask about something he doesn't know a lot about...no problem. He just says up front that he's not too familiar with it, but he's eager to learn about it. That's all he has to say.
Prop him up by saying the hardest part is over. He showed up the first time and did well enough for them to ask him to come back. Having the courage to fight through the shyness to actually get there the first time is harder than actually dealing with the interview. Tell him that you could see the potential even though he wasn't too talkative with you, so they will too.

There's no surefire way to just get over it with a snap of the fingers. He can just mentally prepare.
